The Hybridoma Antibody Screening Kit (FLAG-tagged Antigen, Mouse IgG) is designed for rapid differentiation of antibody affinities in mouse hybridoma cell culture supernatants, enabling fast selection of desired positive hybridoma clones. This kit utilizes KeyTec® TR-FRET technology in a sandwich immunoassay format, offering advantages such as simplicity, speed, high specificity, high sensitivity, and excellent reproducibility.
The basic principle of this method is illustrated below: The KeyTec® TR-FRET Solar Eu1-conjugated anti-FLAG tag antibody binds to the FLAG-tagged antigen; the KeyTec® TR-FRET LA2-conjugated anti-mouse IgG antibody binds to the mouse IgG antibodies expressed by mouse hybridoma cells. When the FLAG-tagged antigen binds to the mouse IgG antibody, the TR-FRET donor and acceptor are brought into close proximity, enabling energy resonance transfer upon external light excitation. By measuring the signal intensity at the specific wavelength (665 nm), the relative affinity of the antibodies expressed by mouse hybridoma cells can be determined.
