KeyTec® TR-FRET cAMP Higher Range Detection Kit can be used to measure cAMP levels in cells. This kit is a competitive immunoassay developed using KeyTec® TR-FRET technology, featuring simple and rapid operation, high specificity, high sensitivity, and excellent reproducibility.
The basic principle of this method is illustrated in the figure below: the specific antibody recognizing cAMP and cAMP are conjugated to KeyTec® TR-FRET Solar Eu and KeyTec® TR-FRET LA, respectively. When mAb anti-cAMP–Solar Eu binds to cAMP–LA, energy resonance transfer occurs between the donor and acceptor upon excitation by an external light source. After cell lysis, intracellular free cAMP is released and competes with cAMP–LA for the binding sites of mAb anti-cAMP–Solar Eu. The signal at the specific wavelength of 665 nm decreases as the concentration of free cAMP in the sample increases.
