The KeyTec® TR-FRET Human Fc Detection Kit is designed for the quantitative detection of human IgG levels in cell culture supernatants or purified samples. This kit utilizes KeyTec® TR-FRET technology in a competitive immunoassay format, offering advantages such as simplicity, speed, high accuracy, excellent reproducibility, and a wide detection range.
The basic principle of this method is illustrated below: The KeyTec® TR-FRET Solar Eu1-conjugated anti-Human Fc specific antibody and the KeyTec® TR-FRET LA2-conjugated Human IgG are used as detection reagents. When the mAb anti-hFc-Solar Eu binds to Human IgG-LA, the TR-FRET donor and acceptor are brought into close proximity, enabling energy resonance transfer upon external light excitation. Free Human IgG present in the sample competes with Human IgG-LA for binding to the mAb anti-hFc-Solar Eu. As a result, the signal intensity at the specific wavelength (665 nm) decreases with increasing concentrations of free Human IgG in the sample.
