KeyTec® TR-FRET Phospho-EGFR (Y1068) Kit can detect the level of intracellular phosphorylated EGFR (Y1068) protein after cell lysis. Epidermal Growth Factor Receptor (EGFR, also known as ErbB-1 or HER1) is a member of the ErbB family of receptor tyrosine kinases. It is located on the cell membrane surface and, upon ligand binding, induces dimerization, which leads to the activation of its intrinsic kinase domain and phosphorylation at specific tyrosine residues. This triggers receptor autophosphorylation and activates downstream signaling pathways, including MAPK, AKT, and JNK pathways, ultimately promoting cell proliferation. EGFR is closely associated with tumor cell proliferation, angiogenesis, tumor invasion and metastasis, and inhibition of apoptosis, making it a critically important therapeutic target in oncology.
The basic principle of this method is illustrated in the figure below. A pair of specific antibodies are conjugated to KeyTec® TR-FRET Solar Eu and KeyTec® TR-FRET LA, respectively: one antibody specifically recognizes the phosphorylation site, while the other recognizes another epitope of the target protein. When the two antibodies simultaneously bind to distinct epitopes of the phosphorylated EGFR (Y1068) protein, the TR-FRET donor and acceptor are brought into close proximity. Upon excitation by an external light source, energy resonance transfer occurs between the donor and acceptor. The level of phosphorylated EGFR (Y1068) protein can then be determined by measuring the signal intensity at a specific wavelength (665 nm).
