The KeyTec® TR‑FRET Revamped TKs Kinase Assay Kit detects the relative activity level of tyrosine kinases (TKs). The kit includes a universal tyrosine kinase substrate and a set of universal detection reagents to determine kinase activity. It can also be used with a variety of tyrosine‑specific substrates optimized by Yaokeyuan. The tyrosine kinase family comprises numerous members, and this kit is compatible with activity detection for over 60 tyrosine kinases (data are continuously updated; please visit our website or contact customer service for details). The kit utilizes an optimized biotinylated peptide as the substrate, which binds to streptavidin‑HX (conjugated with KeyTec® TR‑FRET HX1). Together with an anti‑phospho‑Tyr‑specific antibody labeled with KeyTec® TR‑FRET Solar Eu2, it enables quantification of relative enzyme activity or inhibitor performance by detecting the level of phosphorylated substrate.
This assay is based on a sandwich immunoassay method using KeyTec® TR‑FRET technology, offering a simple, rapid, highly specific, sensitive, and reproducible detection process. The basic steps and principle are illustrated in Figure 1. In the first phase (kinase reaction), the kinase phosphorylates the specific substrate in an optimized reaction buffer containing ATP. In the second phase (detection), the addition of a specific antibody (containing EDTA to terminate the kinase reaction) and streptavidin‑HX allows detection of the phosphorylated substrate. The antibody specifically binds to the phosphorylation site, and streptavidin‑HX binds to the biotin tag on the substrate, bringing the TR‑FRET donor and acceptor into close proximity. Upon excitation by an external light source, resonance energy transfer occurs, generating a TR‑FRET signal. The signal intensity is directly proportional to the amount of phosphorylated substrate.
