Overview

KeyTec® TR-FRET Human STING WT Protein Binding Assay Kit can be used to measure the binding of ligands or inhibitors to wild-type human STING protein. This kit is a competitive immunoassay developed using KeyTec® TR-FRET technology, featuring simplicity, speed, high specificity, high sensitivity, and excellent reproducibility. The kit contains Tag1-labeled wild-type STING protein (stimulator of interferon genes protein, Tag1-STING WT) and a STING WT ligand (an optimized high-affinity ligand) conjugated with KeyTec® TR-FRET Green. It is suitable for high-throughput screening and affinity determination of STING WT inhibitors or ligands.

The basic principle of this method is as follows: the Bright Tb2-conjugated anti-Tag1 antibody specifically recognizes the Tag1-STING WT protein, while the Green1-conjugated STING WT ligand binds to the STING WT protein, bringing Bright Tb and Green into close proximity. Upon excitation by an external light source, energy resonance transfer occurs between the donor and acceptor. Detection of signal intensity at a specific wavelength (520 nm) allows measurement of the binding between the STING WT ligand and the STING WT protein. In the absence of inhibitors or competing ligands in the system, the signal is at its maximum. As the concentration of added inhibitors or ligands increases, the signal gradually decreases. The TR-FRET signal intensity is inversely proportional to the affinity of the inhibitor or ligand.
