The KeyTec® TR-FRET Human CD40/CD40L Binding Assay Kit can be used to measure the interaction between CD40 and CD40L proteins. This kit employs KeyTec® TR-FRET technology and is suitable for high-throughput screening of small-molecule inhibitors, peptides, or antibody blockers, offering simplicity, speed, and high accuracy.
The basic principle of this method is shown in the figure below. KeyTec® TR-FRET Solar Eu1-conjugated Streptavidin (hereafter abbreviated as SA) specifically recognizes biotinylated Human CD40 protein (AVI-Biotin), while KeyTec® TR-FRET LA2-conjugated anti-Tag1 specific antibody specifically recognizes Tag1-tagged Human CD40L protein. When CD40 binds to CD40L, the tag antibodies or affinity reagents conjugated with Solar Eu and LA, respectively, come into proximity; upon excitation by an external light source, resonance energy transfer occurs between the donor and acceptor. Measuring the signal intensity at a specific wavelength (665 nm) allows determination of the extent of interaction between CD40 and CD40L proteins. Therefore, compounds, peptides, or antibodies that block the CD40-CD40L interaction will reduce the TR-FRET signal.
