KeyTec® TR-FRET Human PARG Inhibitor Screening Kit is designed for screening inhibitors of the PARG enzyme. This kit employs an immunoassay developed using KeyTec® TR-FRET technology, featuring simplicity, speed, high accuracy, and excellent reproducibility.
The basic principle of this method is illustrated in the figure below. The KeyTec® TR-FRET Solar Eu1-conjugated anti-Tag1 antibody and the KeyTec® TR-FRET LA2-conjugated Streptavidin (SA) can bind to the Tag1-tagged Biotin-PARylated protein substrate, bringing Solar Eu and LA into close proximity. Upon excitation by an external light source, energy resonance transfer occurs between the donor and acceptor, allowing detection of signal intensity at a specific wavelength (665 nm). The PARG enzyme hydrolyzes the biotinylated poly(ADP-ribose) chains attached to the protein substrate. When the system contains a PARG enzyme inhibitor, the hydrolysis activity of PARG is impeded, and the TR-FRET signal intensity is directly proportional to the inhibitor concentration in the sample.
