Price: $
CAT.: A1090013D
The KeyTec® TR-FRET Human SMARCA2 Protein Binding Assay Kit is designed for screening ligands or inhibitors of wild‑type human SMARCA2 protein. This kit is a competitive immunoassay developed using KeyTec® TR‑FRET technology, offering advantages such as simple and rapid operation, high specificity, high sensitivity, and excellent reproducibility.
The principle is as follows: Solar Eu1‑conjugated anti‑Tag1 antibody specifically recognizes Tag1‑SMARCA2 protein, while HX2‑conjugated streptavidin (Streptavidin‑HX) recognizes biotinylated Bromodomain Ligand. When SMARCA2 protein binds to the Bromodomain Ligand, Solar Eu and HX are brought into close proximity, enabling energy resonance transfer between the donor and acceptor upon external light excitation. The degree of binding between the Bromodomain Ligand and SMARCA2 is determined by measuring the signal intensity at 665 nm. In the absence of inhibitors or competing ligands, the signal is at its maximum; as the concentration of added inhibitors or ligands increases, the signal gradually decreases. The TR‑FRET signal intensity is inversely proportional to the affinity of the inhibitor or ligand.
Standard curve
Validation results with different final concentrations of DMSO
Validation results in buffer systems with different pH values
