The KeyTec® Human TR-FRET IL23/IL12R-β1 Binding Assay Kit is designed for high-throughput screening of small-molecule inhibitors, peptides, or antibodies that block the interaction between IL23 and IL12R-β1 proteins. This kit utilizes KeyTec® TR-FRET technology, offering advantages such as simplicity, speed, and high accuracy.
As illustrated in the principle diagram below, the KeyTec® TR-FRET Solar Eu®¹-labeled anti-Tag 1 antibody specifically recognizes IL23 protein carrying Tag 1, while the KeyTec® TR-FRET LA®²-labeled Streptavidin recognizes biotinylated IL12R-β1 protein (AVI-Biotin). When IL23 binds to IL12R-β1, Solar Eu and LA are brought into close proximity, enabling energy resonance transfer between the donor and acceptor upon external light excitation. The interaction level between IL23 and IL12R-β1 is determined by measuring the signal intensity at a specific wavelength (665 nm). Consequently, compounds, peptides, or antibodies that disrupt the IL23–IL12R-β1 interaction will result in a decrease in TR-FRET signal.
