The KeyTec® TR-FRET Human IL17AA/IL17RA Binding Assay Kit is designed for measuring the interaction between IL17A and IL17RA proteins. Employing KeyTec® TR-FRET technology, this kit is suitable for high‑throughput screening of small‑molecule inhibitors, peptides, or antibody blockers, and is characterized by simplicity, rapidity, and high accuracy.
The basic principle of this method is as follows (as shown in the figure below): KeyTec® TR-FRET Solar Eu‑conjugated Streptavidin and biotinylated Human IL17A protein (AVI‑Biotin) are pre‑bound to form a complex, while KeyTec® TR‑FRET LA‑conjugated IL17RA protein is also provided. When IL17A binds to IL17RA, the respective tag antibodies or affinity reagents conjugated with Solar Eu and LA are brought into close proximity. Upon excitation by an external light source, resonance energy transfer occurs between the donor and the acceptor. Detection of the signal intensity at a specific wavelength (665 nm) allows quantification of the degree of interaction between IL17A and IL17RA proteins. Consequently, compounds, peptides, or antibodies that block the IL17A‑IL17RA interaction will cause a decrease in the TR‑FRET signal.
