The KeyTec® Human TR-FRET IL6/IL6R Binding Assay Kit is designed for high-throughput screening of small-molecule inhibitors, peptides, or antibodies that block the interaction between IL6 and IL6R proteins. This kit utilizes KeyTec® TR-FRET technology, offering advantages such as simplicity, speed, and high accuracy.
As illustrated in the principle diagram below, the KeyTec® TR-FRET Solar Eu®¹-labeled anti-Tag 1 antibody specifically recognizes IL6 protein carrying Tag 1, while the KeyTec® TR-FRET LA®²-labeled Streptavidin recognizes biotinylated IL6R protein (AVI-Biotin). When IL6 binds to IL6R, Solar Eu and LA are brought into close proximity, enabling energy resonance transfer between the donor and acceptor upon external light excitation. The interaction level between IL6 and IL6R is determined by measuring the signal intensity at a specific wavelength (665 nm). Consequently, compounds, peptides, or antibodies that disrupt the IL6–IL6R interaction will result in a decrease in TR-FRET signal.
