The KeyTec® Human TR-FRET IL23/IL23R Binding Assay Kit is designed for high-throughput screening of small-molecule inhibitors, peptides, or antibodies that block the interaction between IL23 and IL23R proteins. This kit utilizes KeyTec® TR-FRET technology, offering advantages such as simplicity, speed, and high accuracy.
As illustrated in the principle diagram below, the KeyTec® TR-FRET Solar Eu®¹-labeled anti-Tag 1 antibody specifically recognizes IL23 protein carrying Tag 1, while the KeyTec® TR-FRET LA®²-labeled Streptavidin recognizes biotinylated IL23R protein (AVI-Biotin). When IL23 binds to IL23R, Solar Eu and LA are brought into close proximity, enabling energy resonance transfer between the donor and acceptor upon external light excitation. The interaction level between IL23 and IL23R is determined by measuring the signal intensity at a specific wavelength (665 nm). Consequently, compounds, peptides, or antibodies that disrupt the IL23–IL23R interaction will result in a decrease in TR-FRET signal.
